You can tighten the clamp onto the phone quite firmly without risk of causing damage to the phone (especially with a protective case). However, do not tighten it as hard as you can as it may damage your phone. Once it has a good grip on the phone it is enough.
For best grip, clamp onto the thickest part of the handlebar available.
